Animals were immunized with mouse splenic or thymic lymphocytes.
Spleen cells were fused with NS-1 myeloma cells.
Can be used in a sandwich killing cytotoxicity assay when conjugated with arsanilic acid and used in conjunction with rabbit anti arsanilic acid antibody.
Tested and found negative for ectromelia virus (mousepox).

Propagation: 
ATCC complete growth medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10.
Atmosphere: air, 95; carbon dioxide (CO2), 5
Temperature: 37.0°C

Subculturing: 
Protocol: Cultures can be maintained by addition of fresh medium or replacement of medium. Adherent cells can be dislodged by scraping. Cultures can be established by centrifugation with subsequent resuspension at 2 to 3 X 10(5) viable cells/ml. A saturation density of 10(6) cells/ml is obtainable.
Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density)

Preservation: 
Freeze medium: Complete growth medium supplemented with 5 (v/v) DMSOComplete growth medium supplemented with 5 (v/v) DMSO
Storage temperature: liquid nitrogen vapor phase
